N-Ethylmaleimide influenced the evaluation of disulfide cross-links in the oxidized myofibrillar proteins using the non-reducing SDS-PAGE

The present study aimed to investigate the effect of N-Ethylmaleimide (NEM) on evaluation disulfide formation in oxi dized myofibrillar proteins during sample preparation non-reducing SDS-PAGE procedure. For this purpose, extracted were oxidized firstly via a Fenton oxidation reaction, and non-oxidized used as control. Before running SDS-PAGE, preparation, these prepared according three dif ferent methods with or without presence β-mercaptoethanol. Results showed that treated NEM regardless presented attenuated bands myosin heavy chain monomer gels, suggesting bonds formed result protein could be preserved by preparation. Meanwhile, possible mechanism for was proposed.
